This is for new primers that we have just received. We get primers delivered in 'blue cap' tube.

  1. Ensure that the primer identity and sequence are in the primer database (http://165.82.245.159:5000/d/s/442067910666494026/474zAg8OIyHRjI9fAxBRJUFx36gI8MWO-UyCAAD3FIgY_).
  2. Turn on a dry bath to 60 degrees.
  3. Write the "O-###" on the cap of the blue tube.
  4. Centrifuge tubes for 30 seconds. This spins down the freeze-dried primers to the bottom on the tube, so that they do not blow out when we open the tube.
  5. Add the appropriate amount of molecular grade water to the tubes for a final concentration of 100uM. The volume can be found on a print-out that came with the primers. It should be around 200ul.
  6. Place primer tubes at 60 degrees for 30-120 minutes.
  7. Vortex
  8. Make your own dilution (typically 1:50 dilution) of this primer in molecular grade water.
  9. Place blue capped tube in the -20 freezer, in one of the primer boxes, in order.
