This is for new primers that we have just received. We get primers delivered in 'blue cap' tube.
- Ensure that the primer identity and sequence are in the primer database (http://165.82.245.159:5000/d/s/442067910666494026/474zAg8OIyHRjI9fAxBRJUFx36gI8MWO-UyCAAD3FIgY_).
- Turn on a dry bath to 60 degrees.
- Write the "O-###" on the cap of the blue tube.
- Centrifuge tubes for 30 seconds. This spins down the freeze-dried primers to the bottom on the tube, so that they do not blow out when we open the tube.
- Add the appropriate amount of molecular grade water to the tubes for a final concentration of 100uM. The volume can be found on a print-out that came with the primers. It should be around 200ul.
- Place primer tubes at 60 degrees for 30-120 minutes.
- Vortex
- Make your own dilution (typically 1:50 dilution) of this primer in molecular grade water.
- Place blue capped tube in the -20 freezer, in one of the primer boxes, in order.
